Cell Culture Media
Cells producing therapeutic proteins require precise nutritional ratios to ensure their replication and growth. These nutrients are provided by the culture media, which must be sterile. If the media contain animal tissue components, terminal sterile filtration should ensure the removal of mycoplasma.
Media are typically prepared in large batches and stored in containers. Before entering the fermentation tank or bioreactor, they are filtered using sterile filtration membranes. Prefiltration is used to remove large particles and colloidal contaminants from the media, extending the lifespan of the terminal sterile filter.
Purpose of Separation
● Prefiltration: To remove lipids, colloids, and particles before terminal filtration.
● Sterile Filtration: To remove bacteria and mycoplasma.
Application Requirements
● Prefiltration should effectively remove particles and colloidal contaminants without impeding the passage of essential media components.
● To achieve timely and cost-effective separation, prefiltration must offer high flow rates and sustained high throughput. Additionally, as the composition of media batches may vary, the designed filtration system should be compatible with these differences.
